Lilith's against Sam right from the start. I'm thinking that she wants to be Lucifer's second-in-command, and Sam's her rival for that position. The YED was in one camp, the one that wants to put Sam as Lucifer's general, while Lilith's in the other camp.
Not sure yet where Dean's in all this. Maybe to reign in Sam perhaps, as perhaps they're seeing Sam as more controllable compared to Lilith, because of Dean. Maybe.
